ABSTRACT

Objective::A new-type electrochemical sensor was developed by electrodepositing gold nanoparticle on the surface of glassy carbon electrode (GCE) modified by Nafion-multi-wall carbon nanotube (MWNTs) by the potentiostatic method, and used for the detection of rutin in practical samples. Method::The electrochemical properties of the sensor in potassium ferricyanide-potassium ferrocyanide system were studied by electrochemical impedance spectroscopy and cyclic voltammetric method, in order to explore the electrochemical performance of the sensor. The electrochemical behavior of rutin on the surface of the sensor was studied by the cyclic voltammetric method, and the factors affecting the performance of the sensor were optimized. The content of rutin was determined by differential pulse voltammetric methods. Result::The optimized conditions were -0.25 V of the deposition potential of gold nanoparticle, 20 s of the deposition time, pH 3.0, and 6 μL of 1 g·L-1 carbon nanotube. Under the conditions, a good linear relationship between reduction peak current and rutin concentration was obtained from 5.0 ×10-9 to 7.0 ×10-7 mol·L-1, and the detection limit was 3.6×10-9 mol·L-1. Conclusion::The developed sensor has a good electrical conductivity and stability, and is highly sensitive and simple for the detection of rutin, with a low detection limit. It has been successfully applied to the detection of rutin in Sophorae Immaturus Flos with the recoveries between 97.6%and 104.4%, which provides a new method for the determination of rutin and a new idea for the quality control of Chinese medicinal materials.

ABSTRACT

Objective: To investigate the influence factors and dynamic varitation rule for the accumulation of huperzine A (Hup A) from thallus of Huperzia serrata, which could produce Hup A. Methods: The optimal concentration of plant growth substances for the growth of thallus of H. serrata were screened using orthogonal design method. The content of target extract from thallus of H. serrata were tested using HPLC. The kinetic curve of thallus growth was described using Logistic growth model. Results: The thallus of H. serrata could produce the most Hup A (72.45 μg/L) when cultured in 1/4 MS solid medium complemented with 12 h/d white light and the relative growth rate could reach 3 174.5%. According to the growth curve of thallus of H. serrata like "S" shape, the best successive generation date of callus culture of H. serrata was 50 d. The concentration of Hup A increased obviously after 60 d when the thallus reached the stable period and the maximal value of Hup A was 7.453 μg/g at 85 d, which meant that the rapid growth of thallus was priority to the accumulation of Hup A. Conclusion: The relational model between the accumulation of Hup A from thallus and its growth is non-growth conjugation. In addition, the growth law of thallus is consistent with Logistic growth model and the maximum specific growth rate (μmax) was 0.071 d-1.

ABSTRACT

Objective To construct a recombinant adenovirus vector carrying hSulf-1 gene and to investigate its effect on the proliferation and migration of human umbilical vein endothelial cells. Methods The hSulf-1 protein was inserted into adenovirus genome to generate recombinant adenovirus Ad5-hSulfl, and then the virus was used to infect ECV-304 cell line. The expression of hSulf-1 protein was detected by Western blotting analysis, the cell viability was examined by MTT assay, and the cell migration ability was evaluated by wound-healing assay in ECV-304 cells. Results The recombinant adenovirus Ad5-hSulfl was successfully constructed. Western blotting analysis showed that over-expression of hSulfl down-regulated the phosphorylation levels of Akt and ERK in ECV-304 cells. MTT assay showed that over-expression of hSulfl inhibited the proliferation of ECV-304 cells, with the cell survival rate decreased to (68. 49 ± 0. 05)% at MOI of 50 pfu/ml and (67. 78 ± 0. 06)% at MOI of 100 pfu/ml(P<0. 05). Wound-healing assay showed that over-expression of hSulfl significantly inhibited the migration of ECV-304 cells compared with the control group (P<0. 01). Conclusion The recombinant adenovirus Ad5-hSulfl-mediated hSulf-1 over-expression can markedly inhibit the proliferation and migration of ECV-304 cells, laying a foundation for hSulf-1 related gene therapy of tumor and angiogenesis-associated diseases.

ABSTRACT

<p><b>OBJECTIVE</b>To introduce the principle, procedure, efficacy and application of SNPstream genotyping technology.</p><p><b>METHODS</b>Genotyping results of 152 SNPs were used to analyze the feasibility, call rate and accuracy of SNPstream technology.</p><p><b>RESULTS</b>For the 152 selected SNPs, 122 SNPs can be genotyped with SNPstream, for which 116 SNPs were successfully genotyped. Replication study showed that the repeatability of genotyping is 99%. When the allele cluster was clear, the accuracy can reach 100%. But when the allele cluster was obscure, the accuracy was only 93.8%.</p><p><b>CONCLUSION</b>SNPstream technology has the advantages of high accuracy, flexible throughput, and high cost performance, and may have a wide application for medical genetics research.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the therapeutic efficacy of interferon (IFN) therapy and risk of long-term administration for chronic hepatitis C (CHC) patients who cannot tolerate the standard treatment.</p><p><b>METHODS</b>Forty-six CHC patients who had proven intolerant to standard treatments were treated with low-dose IFN (non-pegylated IFN: 60 to 300MIU QOD, or pegylated IFN: 50 to 90 mug/w) plus ribavirin (RBV; 0.6g to 0.9 g/d) for 72 weeks.</p><p><b>RESULTS</b>Forty-three (93.5%) of the patients were able to tolerate the long-term treatment with low-dose IFN plus RBV. Only three patients experienced severe side effects (low white blood cell and platelet counts) that required treatment withdrawal. The virology response rates over treatment time were: rapid virologic response (RVR): 10.9%; early virus response (EVR): 30.4%; 24 week virologic response: 45.7%; and, 48 week virologic response: 47.8%. B-sonographic imaging revealed that three patients experienced improved liver morphology through the treatment course. The patients who achieved RVR, EVR, or 24 weeks virologic response also attained higher 48 week virologic response. The 24 week virologic response had the strongest predictive value of good prognosis.</p><p><b>CONCLUSIONS</b>Our study demonstrated that long-term treatment with low-dose interferon plus ribavirin is effective for patients who are otherwise intolerant to standard treatment. In these patients, low-dose IFN plus RBV can obtain a high virologic response rate at 48 week. Furthermore, the 24 week virologic response is sufficiently predictive of treatment success. As with any treatment regimen, it is important for healthcare workers to monitor the disease status and potential side effects throughout the course of therapy.</p>

ABSTRACT

Objective To explore the effects of leukocyte in the injured foci on brain edema after brain contusion,in order to provide theoretical basis of clincial treatment of traumatic brain injury.Methods Traumatic brain injured animal model was established with injecton of blood components,forty five rats were randomly divided into three groups i.e,traumatic brain injure (TBI) group,whole blood(WB)group and white blood cell(WBC) group.Each group was further divided into three subgroups.All rats were killed at 24, 72 or 120 hours after injury,the water content, sodium, potassium and calcium contents were measured in brain tissue of areas injured.Results There was a peak of brain water and sodium contents in TBI, WB and WBC groups at 3 days after injury;Brain water content of TBI group was significantly lower than both WB group and WBC group (P
